The diversion via Nissyros, Tilos and Symi between Kos and Rhodes is subsidised by the government. The final approval for the actual timetable hasn't yet been given by the Ministry of Marine, though there's absolutely no reason to suppose it won't be. It is slightly complicated by the Diagoras having its annual overhaul for the first two weeks of June and being replaced by the Blue Star Paros.
In fact there's no need to book this ferry journey so far in advance. both the ships have a capacity of over 1400 people, and even if it was full leaving Piraeus (unlikely in June), by the time they get to Kos a fair few will have got off already, and not many people will want to get on at 3am.
Nobody travelling all the way from Piraeus to Rhodes would be on this ferry in the first place as the Blue Star2 leaves Piraeus 3 hours later and arrives Rhodes an hour earlier - so the main traffic is trucks and cars.
You could buy tickets in agencies in Kos Town or failing that there's a ticket kiosk next to the passenger waiting shelter on the quay at Kos which will be open half an hour or so before the boat arrives.
